Comment typeDescription
Alternative ProductsEvent=Alternative splicing; Named isoforms=3; Name=1; IsoId=Q9UMR5-1; Sequence=Displayed; Name=2; Synonyms=I; IsoId=Q9UMR5-2; Sequence=VSP_005188; Note=Catalytically inactive due to lack of His-283. May be produced at very low levels due to a premature stop codon in the mRNA, leading to nonsense-mediated mRNA decay.; Name=3; IsoId=Q9UMR5-3; Sequence=VSP_054027; Note=No experimental confirmation available.;
Biophysicochemical PropertiesKinetic parameters: KM=67 uM for S-palmitoyl-CoA ; KM=37 uM for S-palmitoyl-N-acetylcysteamine ; KM=117 uM for 4-methylumbelliferyl-6-S-palmitoyl-beta-D- glucopyranoside ; Vmax=1.7 umol/min/mg enzyme toward S-palmitoyl-CoA ; Vmax=3.3 umol/min/mg enzyme toward S-palmitoyl-N- acetylcysteamine ; Vmax=0.43 umol/min/mg enzyme toward 4-methylumbelliferyl-6-S- palmitoyl-beta-D-glucopyranoside ; pH dependence: Optimum pH is 7.0. ;
CautionWas originally referred as a palmitoyl-protein thioesterase (palmitoyl-protein hydrolase)
FunctionRemoves thioester-linked fatty acyl groups from various substrates including S-palmitoyl-CoA. Has the highest S- thioesterase activity for the acyl groups palmitic and myristic acid followed by other short- and long-chain acyl substrates. However, because of structural constraints, is unable to remove palmitate from peptides or proteins. {ECO:0000269|PubMed:10417332, ECO:0000269|PubMed:12855696, ECO:0000269|PubMed:9341199}.
Sequence CautionSequence=CAG38577.1; Type=Erroneous initiation; Note=Translation N-terminally shortened.; Evidence= ;
SimilarityBelongs to the palmitoyl-protein thioesterase family
Subcellular LocationLysosome
Tissue SpecificityBroadly expressed, with highest levels in skeletal muscle
